What is Agmatine Sulfate?
Agmatine Sulfate, also known as 1-(4-Aminobutyl)guanidine, is naturally derived from a chemical called arginine. It is an ingredient frequently found in a wide range of bodybuilding supplements from pre-workout to post-workout like our intra + post workout supplement: Myocore. Agmatine’s popularity came from its ability to produce nitric oxide (NO), promote muscle growth, improve weight loss, prevent weight gain and much more.
Agmatine is a special supplement because it is a neurotransmitter. It can target receptors in your neurons to help with a range of other diseases and illnesses such as diabetes, Alzheimer’s disease, opioid addiction, mood disorders, and even cancer. Agmatine sulfate also improves your mood and cognitive functions.

Benefits of Agmatine Sulfate Supplementation
Agmatine is most well known for its nitric oxide production (which is why it is found in numerous pre-workout supplements), but an uncommon fact about agmatine sulfate is that it has extremely powerful post workout benefits when supplemented for.
1. Increased Blood Flow (Pump)
- Agmatine’s most well known benefit is its ability to induce muscle pumps. Supplementing with agmatine sulfate pre workout supersaturates the muscles with blood through the modulation of nitric oxide synthesis (NOS). This causes the production of nitric oxide (NO), which will drive blood into the muscle tissue.
- Study Summary: A study tested the effects of agmatine on endothelial cells. They grew a culture of bovine pulmonary artery endothelial cells. When stimulated by agmatine, these cells produced 3x more the basal levels of nitrite. 
2. Promote Muscle Growth
- Agmatine has shown to stimulate the production of luteinizing hormone (LH) in rats. The increase in LH leads to an increase in testosterone levels, which in turn will help create the optimal environment for your body to build muscle and increase performance.
- Study Summary: A study tested the hypothesis that agmatine will stimulate pituitary luteinizing hormone (LH) release. The results imply that the agmatine can serve as an excitatory neurotransmitter to control LH release. 
3. Improve Weight Loss and Prevent Weight Gain
- Agmatine has shown to reduce weight gain, increase fat burning, decrease fat composition and increase muscle mass in rats.
- Study Summary: Agmatine was fed to rats through drinking water for 4 to 8 weeks. It had a widespread impact on the gene expression responsible for metabolism. This includes activating gene expressions to regulate thermogenesis, gluconeogenesis, and carnitine biosynthesis and transport. These drives metabolic changes that went hand in hand with reduced weight gain when the rats were fed an obesity inducing diet. The findings of this study suggests that Agmatine enhances weight loss because it mimics the effects of caloric restriction. 
4. Improve Insulin Sensitivity and Reduce Blood Sugar
- Agmatine sulfate has also shown to increase insulin sensitivity and the uptake of glucose into the muscle. This will allow glucose and amino acids to be sent to the muscles to promote growth and repair, making it a great recovery supplement.
- Study Summary: In a study with mice, the subjects were fed a diabete inducing diet high in fat that is aimed to reduce their insulin sensitivity. Agmatine given to the mice after this diet actually reversed the reduced insulin signalling. Additionally, this study also found the agmatine treatment to have reduced cognitive decline. 
5. Reduction in Pain
- Agmatine has shown to reduce pain in rats with spinal cord injuries and even reduced neuropathic pain by blocking NMDA receptors, which can allow for longer periods of training.
- Study Summary: Injuries in the nerves can cause hypersensitivity to pain and rewire your brain to identify the wrong stimuli as pain signals. Agmatine can suppress your nerves trying to build new but wrong connections. Agmatine has NMDAR antagonist and NOS inhibitor activities that prevent nervous system plasticity (the thing that causes more pain). Not only does it correct hypersensitivity, it also reduces lesion size of the spinal cord injury. 
6. Help with Depression and Anxiety
- Agmatine increases NRF2 in rats, NRF2 protects the brain from high cortisol levels (stress hormone). This helps prevent depressive behaviour in rats.
- Study Summary: In one pilot study containing 3 patients suffering from depression, agmatine supplement helped all 3 patients stop suffering from any more depressive symptoms. The patients refuse to stop agmatine supplements after the study is done because they don’t want to risk relapse. So we do not know the risks of relapse. It is thought that agmatine’s antidepressant effects is caused by blocking NMDA receptors and not through serotonin pathways.

Agmatine Sulfate vs. Arginine
Arginine, an amino acid, has been used for a while now in different forms to promote improved circulation and vascularity through the production of nitric oxide. It can be found in a large variety of cardiovascular health products and majority of pre workout supplements.
Agmatine on the other hand is a recently discovered decarboxylate product of arginine. It shows promising results in also increasing nitric oxide levels that is produced in the human body (amongst other benefits mentioned above).
Agmatine is derived from arginine when you remove the carboxylic acid from the end of the arginine molecule. The resulting molecule can better regulate the production of nitric oxide synthase (NOS), which in turn promote nitric oxide release in your bloodstream. This increases blood blow to your muscles, pumping it with nutrients and oxygen

How Should You Take Agmatine Supplements?
If you take l-arginine, l-citrulline, yohimbine, and/or D-aspartic acid, avoid taking agmatine at the same time as these other supplements. It may hinder agmatine absorption.

Is Agmatine Safe?
A 5 year long case study done on the effect of daily high dosage of Agmatine supplements found evidence that it is safe to consume orally.
Case Summary: Interestingly enough, recent clinical studies have found that it is safer to take agmatine sulfate supplements for 3 weeks than some therapeutics (treatment for neuropathic pain). They have identified dietary agmatine as a nutraceutical.
In this report, human subjects consumed a daily high dosage of oral armatine over a period of 4 to 5 years. The followup physical examinations (including blood and urine analyses) showed that the measurements stayed normal and found no problems. 
Side Effects of Agmatine Sulfate
There are very minor effects of agmatine sulfate, with a very small percentage of people reporting nausea, diarrhea and vomiting. All side effects were cleared after a couple days of discontinued use of agmatine.
How Does Agmatine Work?
While your buddies at the gym may be chatting about how Agmatine gives them pump, how does it really work?
Agmatine being a neuromodulator, modulates enzymes. A neuromodulator is a messenger released from the neuron to control other neurons or cells in your body. You can think of these regulatory compounds in your body as “switches” to control what and how much gets releases. They work synergically to ensure that your body functions optimally. Agmatine is one of these “switches.” It selectively inhibits NOS (nitric oxide synthase). In other words, it stops the production of NOS.
The 3 main types of NOS enzymes are iNOS (inducible), nNOS (neuronal), and eNOS (endothelial). iNOS:
iNOS: makes large amounts of NO to help the immune system kill harmful bacteria. You can think of it as a triggered immune response that can lead to inflammation.
nNOS: it works as a signal between the brain and other neurons. If left unregulated, it can inhibit growth and repair of neurons.
eNOS: signals for vasodilation of blood vessels for increased blood flow. Vasodilation can lead to low blood pressure and improves how fast a cell gets nutrients and get rid of waste products.
Agmatine plays an important role in the regulation process of nitric oxide levels by inhibiting iNOS and nNOS as well as increasing eNOS.
In other words, it reduces inflammation, improve neuron growth and repair, and improves blood flow to your body.
Agmatine controls enzymes in your body to reduce inflammation caused by iNOS, improve neuron growth and repair that would otherwise be limited by nNOS, and improves blood flow to your body by making more of eNOS.
While the exact pathways of Agmatine’s mechanism of action is still under investigation, studies have shown agmatine’s involvements in diabetes (helps it), neuroprotection, and psychiatric conditions.
In diabetes, agmatine helps your body achieve hypoglycemia. Hypoglycemia is a treatment of diabetes because it lowers your blood sugar levels. Agmatine does so by releasing more insulin form pancreatic islet cells. This drives glucose uptake in cells. Agmatine achieves neuroprotection through the modulation of receptors (NMDA, alpha 2, and imidazoline) and ion channels (ATP sensitive potassium channels and voltage-gated calcium channels) as well as blocking nitric oxide synthesis. Agmatine’s inhibition of nNOS helps protech neurons. In terms of psychiatric disorders, agmatine effects mechanism involves neurotransmitter receptor modulation of the NMDA, alpha-2, serotonin, opioid, and imidazoline receptors.
